The Effect of Adriamycin on the Differentiation of Cultured Myoblasts of Chick Embryo

Abstract
This study was performed to investigate the cytotoxicity of Adriamycin on the cultured chicken embryonic skeletal muscle cells. The effect of A-driamycin on the morphological differentiation, cell fusion and the total amount of protein in cultured myoblasts was investigated at higher doses (0.1, 0.5. L pg/ml) and lower doses (0.05. 0.1 pg/ml) of Adriamycin.
The results were as follows :
1. When cultured myoblasts were exposed to lower concentrations of Adriamycin, 0.05pg/ml and 0.1pg/mi, cell proliferation was inhibited partially and cell fusion index was decreased slightly than that of the control group.
Cultured myoblasts were treated with higher concentrations of Adriamycin, 0.1, 0.5 and 1pg/ml for 1 hour and then incubated in Adriamycin -free medium.
Cell proliferation and cell fusion index were similar to those of the control groups in 0.lPg/ml of Adriamycin, and at 0.5pg/ml and Ipg/ml of Adriamycin. Cell proliferation was inhibited slightly but cell fusion index was more increased than that of the control group in myoblasts ex-posed to Adriamycin for 120 hours.
2. In the total amount of protein of myoblasts cultured in lower concentrations of Adriamycin, 0.05pg/ml and 0.lPg-/ml.
The longer the myoblasts exposed to Adriamycin. The more total amount of protein of myoblasts decreased, dose-dependently.
In the total amount of protein of myoblasts exposed to higher concentrations of Adriamycin. 0.1 fig/ml, 0.5 pg/ml and l fig/ml , the total amount of protein in 0.l tag/ml of Adriamycin was similar to that of the control group but at 0.SPg/ml of Adriamycin, the total amount of protein was decreased remarkably than that of the control group.
From the above results it is suggested that Adriamycin decreaes the total amount of protein and inhibits the cell fusion of the cultured myoblasts.
